Research Applications

Assess how patient-derived extracellular matrix changes ovarian-cancer culture behavior and chemotherapy sensitivity.

Model & Design

High-grade serous ovarian cancer PDOs introduced into corresponding decellularized tumor matrix; omentum provides separate matrix characterization material.

Experimental Format

Growth followed to 21 days, with a paired Geltrex comparison in four models. Conventional PDO concentration–response uses 72-hour exposure; the recellularized matrix challenge uses the corresponding PDO-derived IC50.

Model Components

  • Component

    Culture and characterization

    Description

Patient tumor dECM versus Geltrex. DNA depletion, retained collagen/laminin/GAG, collagen structure, proteomics, morphology and Ki67 spatial distribution characterize the systems.

  • Component

    Pharmacology

    Description

Ten-day conventional PDO cultures receive paclitaxel or an equal-ratio paclitaxel–carboplatin combination at the source-reported nominal 1/10/100/1,000 μM series for 72 hours. Recellularized matrix models are challenged with the corresponding PDO-derived IC50. Resazurin fluorescence is measured at 530/590 nm. Drug comparisons use six conventional PDO and five matrix models.

Measurements & Endpoints

  • Growth over 21 days
  • Spatial Ki67 positivity
  • Metabolic activity and fitted IC50
  • Matrix composition and organoid marker agreement

Analysis

Interpret conventional PDO concentration–response, the matrix challenge and separately reported fitted matrix IC50 values as distinct source measurements. Proposed transport or signaling explanations remain unmeasured mechanisms.

Published Evidence

  • Study Sample

Morphology fifteen patient lines, expression nine models, paired growth four, chemotherapy six PDO and five matrix models. These are distinct subsets.

    Comparison

Geltrex PDO culture, corresponding patient tumor dECM constructs and untreated drug controls.

    Finding

Geltrex growth exceeded matrix growth after day six. Paclitaxel IC50 was 37.10 versus 58.8 μM and combination IC50 6.8 versus 27.1 μM in conventional versus matrix cultures. Matrix marginal Ki67 was higher than the whole-construct value.

    Interpretation & Limits
    • Resazurin reports metabolic activity as well as cell survival.
    • Drug penetration was not directly established as the mechanism; growth and signaling may contribute.
    • Decellularization changes the proteome; live stromal/immune cells and paired clinical outcome validation are absent.
    • The main Methods assigns the full nominal concentration series to conventional PDOs and specifies a PDO-derived IC50 challenge for matrix cultures; a matching full matrix series is not specified there.
    Study Design

Primary study comparisons of the stated models, treatments and measurements.
