Research Applications

Test how cancer/stellate background and tissue structure influence aggregate drug response and secreted angiogenic activity.

Model & Design

Human BxPC-3 or MiaPaCa-2 cancer lines, RLT-PSC or hPSC21 stellate lines and HMEC-1 dermal endothelium in defined cell-line cocultures.

Experimental Format

Three-day formation in MCDB131 with 0.24% methylcellulose. Cancer:stellate:endothelial seeding ratios are BxPC-3/RLT-PSC 7:2:4 (5,000 cells), BxPC-3/hPSC21 6:5:3 (7,000), MiaPaCa-2/RLT-PSC 6:3:3 (3,500) and MiaPaCa-2/hPSC21 5:6:4 (4,500). Five-day gemcitabine (2.5 nM–10 μM)/paclitaxel monotherapy and combination assays. The source gives inconsistent paclitaxel:gemcitabine fixed ratios: 1:5 in Methods and 1:8 in Figure 3.

Model Components

  • Component

    Viability

    Description

CellTiter-Glo ATP normalized to untreated and 5 μM staurosporine controls. Response-curve area is used where IC50 cannot be estimated.

  • Component

    Endothelial assay

    Description

Untreated spheroid-conditioned medium and VEGF ELISA; a separate six-hour HMEC-1/Cultrex tube assay uses IKOSA image-analysis software for loops, length, area and branch points.

Measurements & Endpoints

  • Five-day aggregate ATP response-curve area
  • Three-day composition/diameter
  • Conditioned-medium VEGF
  • Six-hour endothelial organization

Analysis

Compare each cancer background with its own monoculture and keep starting cell counts, final fractions and replication units distinct. A conditioned-medium tube assay is separate from vascularization inside the spheroid.

Published Evidence

  • Study Sample

Four defined cocultures. Drug-response measurements n ≥ 7 from three independent experiments; VEGF n = 3 from one experiment; tube assay n ≥ 8 from three experiments.

    Comparison

    BxPC-3 and MiaPaCa-2 monocultures, alternative cancer/stellate pairings, untreated/full-death drug controls and positive/negative tube-assay media.

    Finding

BxPC-3 monocultures were more resistant than their cocultures; MiaPaCa-2 overall sensitivity did not significantly change. BxPC-3/RLT-PSC provided a relatively resistant triple-culture comparison. Conditioned media produced distinct tube area, length and branching patterns without many closed loops.

    Interpretation & Limits
    • The source fixed combination ratio differs between Methods and Figure 3.
    • Different starting cell counts and aggregate ATP prevent cell-type-specific survival inference.
    • HMEC-1 is dermal endothelium; no perfused pancreatic vessel network.
    • No patient-derived or matched clinical treatment validation; the conditioned-media comparison did not test antiangiogenic therapy.
    Study Design

Primary study comparisons of the named models, exposures and measured outcomes.
